Affinity Binding Alginate Biomaterial for the Sustained Delivery of Cardiovascular- Protective Proteins

چکیده
Introduction Myocardial tissue regeneration is an emerging field of interest, aiming to reintroduce cardiac tissue and/or enhance self-repair of the infarct after myocardial infarction (MI). We developed a novel acellular injectable alginate biomaterial by utilizing the ability of alginate-sulfate to affinity-bind and present heparin-binding proteins. We hypothesized that such affinitybinding bioactive hydrogel would potentially induce tissue restoration and self-repair, by expanding the biomaterial effect beyond passive mechanical stabilization of the ventricle. Here, we aimed to test whether the sustained delivery of hepatocyte growth factor (HGF) from such system would enhance its therapeutic properties.

Immunogenic and Protective Potentials of Recombinant Receptor Binding Domain and a C-Terminal Fragment of Clostridium botulinum Neurotoxin Type E
Clostridium Botulinum Type E neurotoxin heavy chain consists of two domains: the translocation domain asthe N-terminal half and the binding domain as the Cterminal half (Hc). One effective way to neutralize botulinum neurotoxin is to inhibit binding of this toxin to neuromuscular synapses with antibodies against binding domain. Two synthetic genes, coding for Hc (the full length binding d...
متن کاملSustained Release of Risedronate from PLGA Microparticles Embedded in Alginate Hydrogel for Treatment of Bony Lesions
Background: Inflammatory bone resorption in periodontitis can lead to tooth loss. Systemic administration of bisphosphonates such as risedronate for preventing bone resorption can cause adverse effects. Alginate hydrogel (ALG) and poly (lactic acid-co-glycolic acid) (PLGA) microparticles have been studied as drug delivery systems for sustained release of drugs. Therefore, the release pattern of...
متن کاملCharacterization and Evaluation of Novel Film Forming Polymer for Drug Delivery
In this study, Damar Batu (DB) − a novel biomaterial− is evaluated for its potential application in pharmaceutical coating. DB is a whitish to yellowish resin, characterized initially in terms of solubility, acid value, molecular weight (Mw), polydispersity index (Mw/Mn) and glass transition temperature (Tg). Neat plasticized films of DB are investigated for mechanical, water vapor transmission...
متن کاملPolyelectrolyte biomaterial interactions provide nanoparticulate carrier for oral insulin delivery.
Nanospheres are being developed for the oral delivery of peptide-based drugs such as insulin. Mucoadhesive, biodegradable, biocompatible, and acid-protective biomaterials are described using a combination of natural polyelectrolytes, with particles formulated through nanoemulsion dispersion followed by triggered in situ gel complexation. Biomaterials meeting these criteria include alginate, dex...
متن کاملIn Silico Design and Verification of LAMP-BDNF Chimeric Protein for Presentation of BDNF on the Surface of Exosomes for Drug Delivery Through Blood-Brain Barrier
Background and purpose: The mature form of brain-derived neurotrophic factor (BDNF) binds to BDNF/NT-3 growth factors receptor (Trk-B). This binding leads to activation of Ras–MAPK pathway which is integrated with cell growth and proliferation. The BDNF deficiency is correlated with various diseases and affects aging and miscellaneous. In the present study we aimed to design a chimeric LAMP-BDN...
